In UE 5.4, the process was to search ‘alpha’ in project settings and change the alpha channel support in postprocessing. In UE 5.5, it’s a bit different when you search for ‘alpha’.
Can someone please explain the (best) process of rendering a transparent background using Movie Render Queue in 5.5 ?

From the UE 5.5 community guidelines, here’s an extract on how to render the alpha channel:
Both .png and .exr image formats offer the ability to output the alpha channel with the image, but first you will need to enable this support. From the editor menu bar, select Edit > Project Settings > Engine > Rendering > Postprocessing > Enable alpha channel support in post processing (experimental) > Linear color space only.
Once this change is applied, you will be prompted to restart the engine. To create a transparent background, you will need to hide opaque objects in your scene such as Sky and Atmospheric Fog.

Unfortunately, the ‘Project Settings’ path you describe is the one from UE5.4. In my UE5.5 project, it is different. Here are some PICS of the differences I am facing.

In UE5.5, I need to know whether I should tick the first box, both boxes, or none, in order to achieve my goal.
After running a few tests, I was able to render a transparent background in UE5.5 by ticking only one box (see picture). In UE5.5, ticking this one box does NOT require a restart.
I took my rendered video to Davinci Resolve and to Adobe Premiere and both allowed me to choose any background image or video I wanted. My rendered video looked like it had a black background but just dropped in and worked in the above programs.
You need to make sure you tick ‘Actor Hidden in Game’ for floor, fog, atmosphere, sky sphere, cloud.
You don’t need to tick any boxes in ‘Deferred Rendering’.
